Malachite green stain

Malachite green stain is a cationic (basic) triphenylmethane dye widely used in microbiology, bacteriology, and cell biology research, primarily for the visualization of bacterial endospores. Because malachite green exhibits a relatively low affinity for vegetative bacterial cells, heat-assisted staining enables the dye to penetrate and become retained within the protective layers of endospores, while it is readily removed from vegetative cells during water decolorization. Subsequent counterstaining, typically with safranin, provides clear differentiation between endospores and vegetative cells. The Schaeffer–Fulton endospore staining method remains the standard laboratory technique for studying sporulation, endospore morphology, and spore distribution in Bacillus, Clostridium, and related genera. In research laboratories, malachite green staining is widely employed in microbial physiology, environmental microbiology, food microbiology, and antimicrobial research to investigate spore formation, persistence, and resistance to physical and chemical stresses. Its simplicity, reproducibility, and compatibility with bright-field microscopy make it a standard method for microbiological research and teaching laboratories.
