Static urine concentrators are innovative ultrafiltration devices designed to enrich low-abundance proteins from urine and other dilute biological fluids prior to diagnostic analyses. Unlike conventional systems, these concentrators operate without centrifugation, enabling clinical laboratories to perform protein enrichment efficiently before electrophoresis or immunoassays. By concentrating dilute samples, they significantly improve the sensitivity of downstream diagnostic techniques.

Passive Concentration Mechanism

These devices rely on a passive ultrafiltration mechanism. Absorbent pads positioned beneath vertically oriented PES membranes create capillary action and osmotic gradients that promote gradual evaporation of water vapor at room temperature. Over a typical period of 30–120 minutes, analytes become concentrated while small molecules such as salts and urea freely permeate the membrane. The dead-stop chamber prevents excessive concentration by halting further volume reduction once the final concentration point is reached. 

Clinical Applications

Static urine concentrators are particularly useful in clinical diagnostics where target proteins are present at very low concentrations. A key application is the detection of Bence Jones proteins (free immunoglobulin light chains) in urine for the diagnosis of disorders such as multiple myeloma and AL amyloidosis using immunofixation electrophoresis (IFE). Because native urine samples often fall below analytical detection limits, pre-concentration improves diagnostic reliability. Additional applications include enrichment of cerebrospinal fluid (CSF) biomarkers in neurodiagnostic studies and enhancement of antibody titers used in bacterial latex agglutination assays for pathogens such as Streptococcus pneumoniae and Haemophilus influenzae. The absence of centrifugation also simplifies workflow and may reduce laboratory costs compared with centrifugal concentration systems.
